干扰素联合反义寡核苷酸体外净化慢粒白血病骨髓

THE EFFECT OF bcr/abl AS-ODN AND IFN IN VITRO BONE MARROW PURGING FOR CHRONIC MYELOGENOUS LEUKEMIA

目的 :探讨干扰素 α2 a、bcr/ abl反义寡核苷酸对慢性粒细胞白血病骨髓体外净化效果。方法 :采用体外克隆形成培养技术 ,分别用于干扰素 α2 a、bcr/ abl反义寡核苷酸不同的组合方案 ,进行体外净化 5例慢性粒细胞白血病 (CML )骨髓。结果 :联用 IFN-α2 a(1× 10 6U / L )、bcr/ abl AS- ODN(4 0 mg/ L )方案 ,对白血病细胞克隆形成单位 (L - CFU )抑制作用有显著差异 (P<0 .0 1) ,而相同条件下 ,GM- CFU及 L - CFU的集落存活率分别为 30 .0 6 %和 8.9% (P<0 .0 1)。结论 :联用 IFN -α2 a(1× 10 6U / L )、bcr/ abl AS- ODN(4 0 mg/ L )方案 ,选择性体外净化慢性粒细胞白血病细胞的效果好 ,可用于临床净化

Objective:To investigate the effect of bcr/abl AS ODN and IFN in vitro bone marrow purging for patients of chronic myelogenous leukemia(CML).Methods:Amplifying in vitro clone formating culture,to purging the bone marrow of 5 CML patients through the different combined regimen among bcr/abl AS ODN and IFN.Results:It has different effects on leukemia cell clone formating unit that combinatin IFN(1×10 6U/L) and bcr/abl AS ODN (40 mg/L).Uder the same ( P <0.01).Conclusion:The combined regimen of IFN(1×10 6U/L) and bcr/abl AS ODN(40 mg/L) has good effect on selective in virto CML bone marrow purging and can be use in clinic purging of CML bone marrow.
