EB病毒BNLF-1基因在恶性血液病的表达及临床意义

EXPRESSION OF EBV-BNLF-1 GENE IN MALIGNANT HEMATOLO-GICAL DISEASE AND ITS CLINICAL SIGNIFICANCE

摘要 目的 :研究 EB病毒 BNL F- 1基因在淋巴瘤和急性白血病的表达及含量 ,分析 T、B细胞性淋巴瘤和急淋、急非淋 BNL F- 1基因表达的差异 ,探讨该基因表达及含量与淋巴瘤患者临床预后的关系。方法 :采用 Am-plisensor荧光定量 PCR技术 ,检测淋巴瘤和急性白血病患者血液或骨髓 EB病毒 BNL F- 1基因片段 ,同时用免疫酶法测定血清中 EB病毒 VCA- Ig A抗体滴度。结果 :(1)淋巴瘤 BNL F- 1基因表达的阳性率高于急淋、急非淋和对照组 ;急淋高于急非淋和对照组 ;急非淋与对照组比较无显著差异。 (2 )淋巴瘤 VCA- Ig A的阳性率高于急非淋和对照组 ;急淋高于对照组 ;急非淋与对照组无显著差异。 (3)淋巴瘤 BNL F- 1基因含量高于急淋、急非淋 ;急淋高于急非淋。 (4 ) T、B细胞性淋巴瘤 BNL F- 1基因表达阳性率及含量无显著差异。 (5 )淋巴瘤 BNL F- 1基因高表达是死亡相关因素之一 ,其含量越高 ,生存期越短。结论 :BNL F- 1基因可能是 EBV的致癌基因之一 ,对淋巴瘤和急淋发的发病起重要作用 ;BNL F- 1基因阳性率及含量的高低可作为淋巴瘤及急淋和急非淋的鉴别诊断依据之一 ;BNL F- 1基因高表达与恶性淋巴瘤的预后呈负相关关系。 Objective:To study the expression and quantity of EBV BNLF 1 gene fragment in lymphoma and acute leukemia and its clinical prognosis in lymphoma.Methods:The BNLF 1 gene fragment was amplified and quantified by using quantitative polymerase chain reaction(PCR),and title VCA IgA was detected by enzyme immunoassay.Results:(1)The positive expressive rate of BNLF 1 was higher in lymphoma than that in acute lymphocytic leukemia,acute non lymphocytic leukemia and the control.There were no significantly different with acute non lymphocytic leukemia and the control.(2)The positive rate of VCA IgA was higher in lymphoma than that in acute non lymphocytic leukemia and the control.It was high acute non lymphocytic leukemia than that in the control.(3)The quantity of BNLF 1 was higher in lymphoma than that in acute leukemia and acute non lymphocytic.It was higher in acute lymphocytic leukemia than that in acute non lymphocytic leukemia.(4)There were no significantly different with T cell lymphoma and B cell lymphoma of expressive rate and quantity of BNLF 1 gene.(5)It showed negative correlation between the quantity of BNLF 1 gene and life span in lymphoma.Conclusion:BNLF 1 may be one of carcinogens to lymphoma and acute lymphocytic leukemia.BNLF 1 may serve as one of the basis of differentiated diagnosis of lymphoma,acute lymphocytic leukemia and acute non lymphocytic leukemia.EBV may involved in T cell lymphoma in addition to involving in B cell lymphoma.BNLF 1 gene is the death danger factor in lymphoma.The higher quantity of BNLF 1 gene,the shorter life span was.
