摘要
本实验应用淋巴细胞杂交瘤技术,建立了分泌抗布鲁氏菌弱毒疫苗M5(或M5—90)菌株的株系特异的单克隆抗体(McAb),杂交瘤细胞株(IC—11)。以其分泌的IC—11McAb的酶结合物,M5菌株特异抗原(P_2)和受检血清建立的免疫班点试验(DB),具有特异地识别出动物出动物接种M5或M5—90疫苗血清抗体能力,其DB反应阴性的血清,再作凝集试验呈阳性为自然感染动物,本法适(?)用布氏菌M5或M5—90疫苗免疫予防地区布病的发病与疫苗接种动物的鉴别,经若干(?),对2573份动物检测,证实有良好的实用价值。
A hybridoma cell line (Jc-11) .which secretes a strain specific monoclonal antibody (McAb) against Brucella Vaccine strain MS (or M5 - 90), was established by lymphocyte hybridoma technique. Afterwards,a immunoblot (IB)test employing IC-llMcAb-horsera-dish peroxidase (HRP)conjugate and strain M5 antigen (P2)wasdeveloped8The serum antibodies from strain M5 or M5 -90noated aninals can be recognized (posi-tive)by IB test. On the other hand,naturally infected animals can be detected by coagulation test from whose sera are negative in IB test. There-fore, the method combining IB test and coagulation test is suitable for distinction Brucella infected animals and vaccinated ones in M5 or M5 - 90 vaccine strain immunized area. In the diagnostic tests for 2753 animals by several stations of veterinary medicines, the method has been demonstrated to be very practical.
