摘要
本文报告了本实验室设计的由血红细胞自溶液60℃热变性, 乙醇——氯仿法除血红蛋白,旋转蒸发法减压浓缩抽去氯仿、乙醇,硫酸铵分级盐析法沉降SOD,Sepbadex G-75层析提纯SOD等步骤构成的一条成本低、设计合理、简便实用的分离纯化SOD的工艺路线。
An improved technology for preparation of superoxide dismutase (SOD) from pig or cattle erythrocytes is presented in this paper. The designed procedures cover (1) 60℃ heat denaturation of autolyzed erythrocytes, (2) removing of hemoglobin with ethanol-chloroform mixture, (3) concentation through rotary vacuum evaporating, (4) precipitation of SOD by (NH4)2SO4 fractino-al salting out and (5) purification of SOD by Sephadex G-75 gel filtration column chromatography.The purified SOD has an UV-spectrum with a maxi-mum absorption at 260 nm. The results indicated that the technological route is rational, practical and of low cost.
