摘要
目的 :探讨药物渗透性与卵巢癌SKOV3ip1细胞形成的多细胞团簇化疗耐药性的关系。方法 :用液体重叠系统获得SKOV3ip1多细胞团簇 ,毛细管区带电泳法测定SKOV3ip1单层和团簇细胞胞内顺铂和紫杉醇含量。结果 :顺铂作用 2h后单层细胞和团族细胞内顺铂浓度分别为 19.2 196± 5 .3111nmol/ (L·10 6细胞 )和 2 1.994 4± 8.9776nmol/ (L·10 6细胞 ) ,差异无显著性 (P =0 .6 3)。紫杉醇作用 1h后单层细胞和团簇细胞胞内紫杉醇浓度分别为 5 .332 9± 0 .9818nmol/ (L·10 6细胞 )和 11.15 99± 0 .70 4 1nmol/ (L·10 6细胞 ) ,差异有高度显著性 (P <0 .0 1)。结论 :同样条件下 ,团簇细胞胞内顺铂和紫杉醇浓度不比单层细胞内药物浓度低 ,顺铂、紫杉醇在卵巢癌SKOV3ip1多细胞团簇中的渗透因素不是引起团簇细胞化疗耐药的原因。
Objective:To detect intracellular concentration of cisplatin and taxol in multicellular aggregates of ovarian cancer SKOV3ip1 cells and investigate the relationship between multicellular mediated drug resistance and drug permeability.Methods:Liquid overlay system was employed to obtain multicellular aggregates.Concentration of cisplatin and taxol was detected with capillary zone electrophoresis.Results: Intracellular concentration of cisplatin in multicellular aggregates and monolayer cells was 19.2196±5.3111nmol/(L·10 6 cells) and 21.9944±8.7996nmol/(L·10 6 cells)respectively.No difference of cisplatin concentration existed in monolayer and aggregates cells after cisplatin exposure for 2 hours (P=0.63).Taxol concentration of aggregates cells was 11.1599±0.7041nmol/(L·10 6 cells) and markedly elevated than monolayer cells[5.3329±0.9818nmol/(L·10 6 cells)](P<0.01). Conclusions:The intracellular concentration of cisplatin or taxol in multicllular aggregates is not less than that in monolayer cells.Drug permeability is not the cause of multicellular mediated drug resistance in multicellular aggregates.
