摘要
本文探讨了基本上属于两态的细胞膜离子通道事件的自动分析方法.对信号采集所涉及的信号滤波,采样时钟和负延时等条件设定,提出了最佳选择方案.对通道事件的加工和主要参数计算,提出了简而易行的数据滤波,基线恢复和伪迹抑制等方法,特别对通道事件的寻找提出了比半幅度阈值法更具优越性的三阈值法.应用以上概念和方法完成了PCLAMP10.7膜片钳制数据处理系统.实践表明:通道开启时间,电流幅度的测量精度以及亚型通道的分辨能力均达到相当可靠的程度,在排除噪声和干扰的影响以及单通道事件的纯化方面,本系统亦具独特功能.
From the two-state events of ion-channel we have approached to a way of automatic analysis technique. Optimum parameters of analog filtering, sampling clock, and negative delay involved in signal sampling were put forward. Data filtering, baseline restoration and artifact cancel in events processing and calculating of primary parameters were discussed in detial. Especially, as for finding channel events, instead of half-amplitude threshold technique, we advanced the three-threshold method which was more advantages than the half-amplitude threshold technique. The data processing system for membrane patch clamping was programmed according to the concepts and methods above. The results shown that measuring accuracy of single-channel open times, current and resolution ability of Ach receptor subtype channel in Xenopus were all reached to a creditable level. The system had also fantastic functions in noise rejection and purification of single-channel events.
