肌浆网钙泵在心房颤动患者心房肌中的表达

THE ROLE OF SARCOPLASMIC RETICULUM CALCIUM ADENOSINE TRIPHOSPHATASE IN CHRONIC ATRIAL FIBRILLATION

摘要 目的 :研究肌浆网钙泵在调节心房颤动 (AF)患者心房肌细胞内Ca2 + 稳态中的作用。方法 :慢性AF患者 10例 (AF组 ) ,对照组 6例。采用三电极直流等离子体原子发射光电直读光谱法测定二级心房肌细胞内Ca2 + 含量 ,采用免疫组化和Westernblot技术测定心房肌肌浆网 (SR)钙泵 (Ca2 + ATPase)蛋白质表达的变化。结果 :AF组心房肌细胞内Ca2 + 含量较对照组高 4 4倍以上〔(132 9 6 3± 796 5 7) μg/mlvs(30 1.6 7± 30 .89) μg/ml,P <0 0 1〕 ;SRCa2 + ATPase蛋白质的相对含量明显低于对照组 (0 74± 0 0 7vs1 0 2± 0 0 4 ,P <0 0 5 )。结论 :慢性AF患者心房肌细胞内Ca2 + 超载 ,SRCa2 + ATPase功能减低是细胞内Ca2 + 超负荷的重要因素。 Objective:To evaluate the role of sarcoplasmic reticulum adenosine triphosphatase (SR Ca 2+-ATPase) in patients with chronic atrial fibrillation (AF).Methods:10 patients with AF and 6 control subjects were included.The content of intracellular calcium in the atrial tissue was assayed by spectrocomparator.The protein levels of SR Ca 2+-ATPase was analyzed by Westem blot analysis and immunohistochemistry.Results:Compared with control subjects,the Ca 2+ concentration was significantly increased 〔(1329.63±796.57)μg/ml vs (301.67±30.89)μg/ml, P<0.01〕 and the protein level of SR Ca 2+-ATPase was reduced (0.74±0.07 vs 1.02±0.4, P<0.05) in patients with AF.Conclusion:The intracellular calcium overload may be an important mediator of AF.Down regulated protein level of SR Ca 2+-ATPase play an important role in intracellular calcium overload.
