摘要
目的 了解口腔鳞癌及一些口腔正常组织的内源性脂肪酸合成活性。方法 切取口腔鳞癌患者新鲜手术标本的癌组织、癌周组织及正常口腔组织块 ,剪成细块 ,与 [U - 14 C]乙酸钠孵育 2 .5h ,提取脂质 ,液体闪烁计数14 C的掺入量。结果 癌组织14 C的掺入量为 395cpm/mg± 146cpm/mg湿组织 ,癌旁为 2 2cpm/mg± 10cpm/mg湿组织 ,口腔粘膜为 176cpm/mg± 6 4cpm/mg湿组织 ,骨骼肌为 7cpm/mg± 2cpm/mg湿组织 ,腮腺为 94cpm/mg±18cpm/mg湿组织 ,脂肪为 16cpm/mg± 5cpm/mg湿组织 ,淋巴结为 12 5cpm/mg± 47cpm/mg湿组织。
Objective Endogenous fatty acid synthetic activities of squamous cell carcinoma and some normal oral tissues were assayed.Methods Squamous cell carcinoma,adjacent tissue,and some normal oral tissues were collected fresh from surgical specimens.The collected tissues were minced in RPMI and incubated in [U- 14 C] acetic acid,sodium salt for 2.5 hours at 37℃ in 5% CO 2.After labeling,total lipids were extracted and counted for 14 C by scintillation counting.Results 14 C incorporation in the total lipids of squamous cell carcinoma was significantly higher than the other tissues.Conclusion Fatty acid synthetic activity in squamous cell carcinoma is much higher than in adjacent and normal tissues.
