口腔鳞癌临床因素与APC基因杂合缺失的研究

Clinical factors and loss of heterozygosity of APC suppressor gene in oral squamous cell carcinoma

目的 分析APC基因杂合缺失与口腔鳞癌病理分级、临床分期之间的关系 ,探索抑癌基因在口腔鳞癌发生、发展中的作用。方法 应用聚合酶链反应限制性片段长度多态性分析方法 (PCR -RFLP)对 40例口腔鳞癌组织中APC基因的杂合缺失 (LOH)进行检测。结果 APC基因的LOH频率为 6 6 .6 % ;病理分级中 ,低、中分化组APC基因的LOH频率均高于高分化组 (P <0 .0 5 ) ;临床分期中 ,APC基因的LOH频率在Ⅰ～Ⅱ期和Ⅲ～Ⅳ期两组间无差异 (P >0 .0 5 )。结论 APC基因的失活在口腔鳞癌的发生。

Objective To analyze the relation between loss of heterozygosity of APC suppressor gene and clinical factors in oral squamous cell carcinoma and to find the role of APC suppressor gene in the development and progression in oral malignant tumors.Methods We used the polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) to examine 40 oral squamous cell carcinomas for loss of heterozygosity (LOH) at APC.Results The LOH at tumor tissues was detected in 66.6% of APC;LOH at APC was correlated with the tumor of histologic grades (P< 0.05 ).However,no significant difference of LOH was found among clinical stages (P> 0.05 ).Conclusion The LOH at APC suppressor gene is an important factor in the development and progression of oral malignant tumors.
