摘要
目前在MEOR研究、菌液检测、现场试验监控及环保领域 ,石油烃降解菌 (HDB)菌数的测定基本上采用费时费事的最大概率数法 (MPN法 )。本文推荐使用简单易行的细菌瓶法。研制了盛有唯一碳源石油烃、氮源、磷源及专用生长指示剂的培养液 ,适用于HDB菌的测试瓶。欲测水样注入测试瓶逐级稀释 ,在 30℃培养 5— 7天。试液由红变黄表示有HDB菌生长。HDB菌的计数按常规细菌瓶法进行。用所推荐的细菌瓶法测定了某油田采出液、污水及采油用菌液中HDB菌数 ,所得结果与MPN法基本相同。HDB菌测试瓶已有工业产品 (HDB JH型 )。
In the researches, bacterial culture inspection and field trial monitoring of MEOR and in invironmental protection, the main method of determining bacterial cell population for hydrocarbon degrading bacteria(HDB) used today is still time consuming and troublesome the most probable number(MPN) tests. For this a simple and convenient culture flask test is recommended. The flasks containing hydrocarbons (as only carbon source) and proper nitrogen and phosphore sources in cultures as well as a special indicator for growth of HDB bacteria. The aqueous samples under test are injected into a series of flasks and diluted successively, step by step, and cultivated at 30℃ for 5-7 days. The change of color of test samples from red to yellow denotes the growth of HDB bacteria. The bacterial cell population is determined by the same procedures as in common flask test. By using the recommended flask test, the bacterial cell population of HDB bacteria in an oilfield produced fluid and an oily sewege, and a bacteria culture for microbial oil recovery at an oil field is determined and practically the same results are obtained as compared with that by MNP test. The culture flasks for HDB bacteria are now manufactured as commercial articles of type HDB JH.
