摘要
目的 探讨无机汞对雄性生殖系统的损伤机制。方法 雄性Wistar大鼠随机分 3组 ,分别隔日皮下注射剂量为 1.0mg/kg、3.0mg/kg的HgCl2 及生理盐水 ,连续 2 1d。观测睾丸及附睾脏器系数、生精细胞凋亡情况、脂质过氧化物 (LPO)、一氧化氮 (NO)含量以及染毒前后血清睾酮水平。结果 (1) 3.0mg/kgHgCl2 染毒组大鼠睾丸及附睾脏器系数分别为 0 .74、0 .2 6 ,均低于对照组(睾丸 0 .89,附睾 0 .30 ) ,差异有显著性 (P <0 .0 5 )。 (2 ) 1.0mg/kg及 3.0mg/kgHgCl2 染毒组染毒后睾酮水平分别为 0 .93、0 .6 9nmol/L ,均低于染毒前 (5 .0 1、3.0 2nmol/L)及对照组 (5 .4 9nmol/L) ,差异有显著性 (P <0 .0 5 )。 (3) 1.0mg/kg和 3.0mg/kgHgCl2 染毒组LPO分别为 17.34、2 1.5 9μmol/ml;NO分别为 6 2 .91、73.0 9μmol/ml,均高于对照组 (LPO 10 .32 μmol/ml,NO 4 9.86 μmol/ml) ,差异有显著性 (P <0 .0 5 )。 (4) 3mg/kgHgCl2 染毒组平均曲细精管面积为 5 6 15 9.0 1μm2 /个 ,低于对照组 (12 2 2 2 74 .39μm2 /个 ) ,差异有显著性 (P <0 .0 5 )。 1.0mg/kgHgCl2 染毒组平均每个曲细精管的凋亡细胞数为 114 .2 0个 ,高于对照组 (39.0 2个 ) ,差异有显著性 (P <0 .0 5 )。 1、3.0mg/kgHgCl2
Objective To explore the injuring mechanism of reproduction system in male rat exposed to inorganic mercury. Methods The male Wistar rats were divided randomly into three groups.1.0 mg/kg,3.0 mg/kg HgCl 2 and physiological saline were given separately by subcutaneous injection every other day for 21 d.The measured items included the organ coefficients of testis and epididymis,the apoptosis of the spermatogenic cells,the contents of LPO,NO and the serum testosterone.Results (1)In 3.0 mg/kg HgCl 2 group,the organ coefficients of testis(0.74) and epididymis (0.26) were lower than that of the controls[testis 0.89;epididymis 0.30( P <0.05)].(2)After the exposure,the serum testosterone levels in two exposed groups were lower(1.0 mg/kg,0.93 nmol/L;3.0 mg/kg,0.69 nmol/L) than that of the control(5.49 nmol/L) and that before exposure(5.21,3.02 nmol/L).There were significant differences, P < 0.05 .(3)In two exposure groups,the levels of LPO and NO were significantly higher(LPO:1.0 mg/kg 17.34 μmol/ml,3.0 mg/kg 21.59 μmol/ml;NO:1.0 mg/kg 62.91 μmol/ml,3.0 mg/kg 73.09 μmol/ml) than that of the controls(LPO:10.32 μmol/ml,NO:49.86 μmol/ml), P <0.05.(4)The average area of seminiferous tubes in 3.0 mg/kg dosage was 56 159.01 μm 2,significantly lower than that of the control ( 1 222 274.39 μm 2), P <0.05.In 1.0 mg/kg dosage,the apoptotic cells per tube were 114.20 on average,significantly higher than that of the control(39.02), P <0.05.The apoptotic cells per unit area of seminiferous were significantly higher(1.0 mg/kg 1.06,3.0 mg/kg 1.04) than that of the control(0.33), P <0.05. Conclusion HgCl 2 might induce apoptosis of the spermatogenic cells.This perhaps was the reaction to the shortage of testosterone.Increased apoptosis of the spermatogenic cells and reduced testosterone might be relative to oxidative stress of the testis.
