摘要
目的 用随机扩增多态 DNA(Random amplified polymorphic DNA,RAPD)方法区分10个常用小鼠品系。方法 从120条随机引物中筛选出稳定性、重复性好的4条多态性引物对10个常用小鼠品系进行RAPD扩增。扩增产物在含溴化乙锭的1.5%琼脂糖凝胶中电泳,紫外透射仪上观察照相。结果 仅用这4条引物就可将10个品系小鼠完全区分开来,其中包括遗传背景或外观形态根相似的品系,如 BALB/c与 BALB/c-un、KM与BALB/c也能区分出来。结论 筛选出的4条引物对实际工作具有指导意义,适用于小鼠常规遗传监测。
Objective To distingush 10 common strains of mice by random amplified polymorphic DNA (RAPD) assay. Methods Four stable and reproducible polymorphic primers screening from set of 120 random primers were used for amplifing the genomic DNA of 10 strains of nice. The amplified products were electrophorised in agarose gel containing and obsersed under UV examiner. Results The the commonly used experimental strains of mice could be distinguished from each other, including the strains similar in genetic background and appearance by the 4 selected primere, such as BALB/c and BALB/c-nu, KM and BALB/c. Conclusion This method be used for mouse genetic monitoring and strain identification.
