摘要
目的 探讨普鲁卡因对心肌细胞缺氧损伤的影响。方法 将原代培养成活 4 8h的大鼠乳鼠心肌细胞分为三组 :A组为对照组 (氰化钠造成心肌细胞细胞内缺氧模型 ) ,B组为Pro1组 (缺氧 +7 4mg/L普鲁卡因 ) ,C组为Pro2组 (缺氧 +14 8mg/L普鲁卡因 )。比较三组心肌细胞细胞形态学 (倒置相差显微镜、透射电镜观察 )的变化及吸光度A值的改变。 结果 缺氧 12h后 ,对照组细胞搏动功能变化明显 ,呈散在细胞搏动 ,而实验组搏动频率减慢。随着时间的延长 ,细胞形态学变化逐渐明显 ,对照组较实验组变化更显著。
Objective To explore the protective effects of procaine against anoxic damages in cell cultured neonatal rat myocardium. Methods The hypoxic damages of cultured myocardial cells in 1~3 day old SD rats were induced with NaCN. The myocardial cells were contineuosly cultured in the culture medium with or without procaine, which were divided into 3 groups, group A as control (hypoxic damages), groups Pro1 and Pro2 treated with procaine (7 4 mg/L,14 8 mg/L respectively) for 2 h to 24 h. The cell morphology A value (cell survival number) of cells were measured in the groups Pro1, Pro2 and the control group respectively. Results After 12 hours of anoxia, in the control group the beating rates of cultured neonatal myocardial cells showed partly beating or no beating, while the cells in the groups Pro1 and Pro2 became slow. The insult effects of anoxia on morphology were obviously showed in a time-dependent manner, the changes were more significant in the control group than in the Pro1 group and Pro2 group. After 24 hours of anoxia, the A value of the control group was decreased from 0 369±0 036 before anoxia to 0 205±0 013. Conclusion Propofol can protect the cultured myocardial cells of neonatal rats against anoxic damages.
