nm23-h1基因导入对腺样囊性癌分化的影响 被引量:6

Effects on differentiation of adnoid cystic carcinoma(ACC) cell line after transfection of nm23-h1 plasmid

摘要 目的 探讨nm23-h1 基因对腺样囊性癌细胞分化的影响。方法 体外用阳离子脂质体介导nm23-h1质粒转染人口腔涎腺腺样囊性癌(ACC)细胞株,筛选出转染阳性细胞克隆,并植入4只棵鼠颌面部,3w后获取标本,石蜡包埋后进行常规组织学及免疫组织化学检查,实验以未转染的阴性细胞克隆植入4只裸鼠作对照。结果nm23-h1阳性ACC植入棵鼠体内后肿瘤细胞有向原代细胞回归的倾向,即由实体型向筛状型或管状型转变,肿瘤细胞周围的肿瘤间质增多,巢状的肿瘤细胞外围有基底膜样结构形成;肿瘤细胞在体内增殖的过程中,表达nm23-h1的细胞数量有逐渐减少、表达强度减弱的趋势。结论 以nm23-h1为靶基因的基因治疗因同时具有抑制转移和促进分化的作用,具有较大的研究价值和临床应用前景。 Objective To investigate the effect of ntn23-hl on the differentiation of adnoid cystic carcinoma( ACC) cell line. Methods After transfection of nnr23-h1 to ACC-M cell line, the positive nrn23-hl clones were selected and transplanted to 4 BALB/c nu/nu mice in the maxillofacial region while another 4 nu/nu mice were transplanted with nm23-hl negatively-expressed clones as control. 3 weeks later, the mice were killed and the tumor samples were gained, HE stains and immunohistological chemistry stains were performed after routine fixation and paraffin-bedding. Results nm23-hl positive-expressed ACC-M had the tendency to differentiate to the phenotype from which it originated, that was tubulin or acinar-liked structures formed during the devolepment of the tumor,extrcellular matrix increase around the acinus, the number of nm23-h1 positive cells arid the strongness of the nm23-hl expression decreased as time goes on. Conclusion nm23-hl gene has prosperous clinical use and needs for study because of its double functions: antimetastasis and promoting differentiation.
