摘要
目的:建立法莫替丁血药浓度测定方法。方法:采用固相萃取高效液相色谱法。样品先经Waters Oasis HLB固相萃取小柱处理。色谱柱:Shim-pack C_8 150mm×4.6mm,10μm;柱温:35℃;流动相为乙腈-0.019mol·L^(-1)磷酸(10:90);流速:1mL·min^(-1);检测波长:265nm。结果:线性范围5~200μg·L^(-1),线性关系良好,最小可测定浓度为5μg·L^(-1)。回收率80%以上,日内及日间RSD小于10%。结论:采用新型固相萃取柱能快速可靠地测定法莫替丁血药浓度。
Objective: To establish a HPLC with solid phase extraction method for determination of famotidine concentration in plasma. Methods: Drug in plasma was extracted with Oasis HLB extraction cartridges the reversed - phase system consisting of Shim - pack C8 column and acetonitrile - 0.019 mol·L-1 phosphoric acid (10:90) , the eluated with a flow rate of 1 mL· min-1, detection wavelength was 265 nm, column temperature was 35℃. Results:: The calibration curves were liner between 5-200 μg·L-1, detectable concentration limited was 5μg·L-1, the recovery rate were about 80% , the precision (RSD) within day and between day were below 10 percent. Conclusion: Solid phase extraction - HPLC method was simple, accurate and suitable for plasma famotidine concentration analysis and its pharma-cokinetics study.
