摘要
目的 研究不同剂量X射线全身照射对小鼠派伊氏板细胞凋亡的影响及作用机理。方法 采用反转录PCR法检测了派伊氏板细胞Bcl 2转录水平的变化 ,并用琼脂糖凝胶电泳、流式细胞术检测了不同剂量X射线全身照射后小鼠派伊氏板细胞DNA及凋亡小体的改变。结果 2GyX射线全身照射使派伊氏板细胞凋亡增多 ,Bcl 2转录水平表达下降 ;而 75mGyX射线全身照射使派伊氏板细胞凋亡减少 ,同时Bcl 2转录水平表达升高。结论 Bcl 2参与了辐射诱导小鼠派伊氏板细胞凋亡的基本调控。
Objective\ To study the effect of whole body irradiation (WBI) with different doses of X\|rays on apoptosis in cells of mouse Peyers patches and its molecular mechanism. Methods\ RT\|PCR was used to detect the changes of Bcl\|2 transcription level.Agarose electrophoresis and flow cytometry were used to detect the changes of DNA and apoptotic bodies in Peyers patches after WBI with different doses of X\|rays. Results\ The apoptotic was increased and Bcl\|2 transcription level was decreased in Peyers patches after 2 Gy X\|rays.The apoptotic rate was decreased and Bcl\|2 transcription level was increased in Peyers patches after 75 mGy X\|rays. Conclusion\ Bcl\|2 participates in the regulation of radiation\|induced apoptosis in Peyer's patches.\;
