培多普利对自发性高血压大鼠心室重塑过程中左室心肌细胞凋亡和Bcl_2、Bax基因表达的影响

摘要 为研究培多普利对自发性高血压大鼠 (SHR)左室心肌细胞凋亡和 Bcl2 、Bax基因表达的影响 ,采用 TUNEL技术及 DNA电泳研究心肌细胞凋亡 ;原位杂交研究 Bcl2 、Bax基因表达。结果显示 :培多普利 1抑制左室心肌细胞凋亡发生 ,2使 Bcl2 / Bax基因表达比率恢复正常。提示 :培多普利使 SHR心肌 Bcl2 / Bax表达比率恢复正常是其抑制心室重塑过程中心肌细胞凋亡的重要机制。 The effects of Peridopril on the cardiac myocyte apoptosis and Bcl 2 , Bax gene expression of spontaneous hypertension rat (SHR) were investigated. Tdt-mediated dUTP nick end labeling (TUNEL) method and DNA electrophoresis were used to study cardiac myocyte apoptosis. In situ hybridization was used to detect Bcl 2 , Bax gene expression in left ventricular cardiac myocyte. The results showed that Peridopril decreased the incidence of cardiac myocyte apoptosis of SHR; Peridopril restored the ratio of Bcl 2 /Bax gene expression. It was concluded that Peridopril restoring Bcl 2 /Bax gene expression ratio contributes to its effect of reducing cardiac myocyte apoptosis incidence during ventricular remodeling of SHR.
