摘要 目的 研究血管内皮生长因子 (VEGF)对脑血管通透性的影响 ,并观察丹参水溶性成份丹酚酸B拮抗VEGF的作用。方法 动脉及侧脑室注射VEGF后 ,再注射伊文思兰 ,用分光光度法测脑内伊文思兰染色程度来反映VEGF的血管通透作用。注射VEGF前 30min ,动脉或侧脑室给药 ,观察药物抑制作用。结果 动脉及侧脑室注射VEGF剂量依赖性引起脑血管通透性升高 ,但两种给药方法无显著性差异。静脉注射丹酚酸B 0 .1mg·kg-1体重可显著抑制VEGF的通透作用 ,而侧脑室注射无明显作用。结论 丹酚酸B可显著抑制VEGF的血管通透作用 。 Aim To examine the effect of recombinant human vascular endothelial growth factor (VEGF) on the permeability of rat brain and the inhibitory effect of salvianolic acid B in vivo.Methods The action of VEGF in the rat brain was determined by using ultraviolet spectrophotometry to examine Evans blue staining after injection of VEGF into rat lateral ventricle or into femoral artery. The drug inhibitory effect was observed by injected salvianolic acid B into vein or lateral ventricle 30 minutes before the VEGF injection.Results Injection of VEGF into rat lateral ventricle or into femoral artery dose-dependently increased the Evans blue staining of the brain,but there was no significantly difference between them. Salvianolic acid B intravenous injection (0.1mg·kg -1 ) could markedly inhibit the permeability effect of VEGF, however lateral ventricle injection had no effect.Conclusion These results suggested that salvianolic acid B could significantly inhibit VEGF-induced hyperpermeability in rat brain, and had certain protection on rat blood vessel of brain.
