摘要
目的研究脑缺血再灌流后海马齿状回神经元发生水平的动态变化及外源性碱性成纤维细胞生长因子(bFGF)对其的影响。方法采用5-溴脱氧尿核苷(5-bromodeoxyuridine,BrdU)标记有丝分裂、免疫组织化学的方法观察大鼠全脑缺血再灌流后海马齿状回神经元发生的动态变化以及在侧脑室注射外源性bFGF对神经元发生变化的影响。结果大鼠全脑缺血-再灌注损伤后第14天齿状回神经元发生水平达到峰值。全脑缺血-再灌注损伤大鼠侧脑室注射外源性bFGF后齿状回神经元发生水平升高,神经元发生峰值时间提前。结论脑缺血可以诱导齿状回神经元发生水平上调,外源性bFGF能促进齿状回神经元发生。
Objective To explore the change of neurogenesis in the dentate gyrus of rats and the e ffect to exogenous bFGF on it after global ischemia reperfusion.Methods Bromodeoxyuridine(BrdU),a mitotic indicator,immunity histochemistry were used to observe the c hange of neurogenesis in hippocampus dentate gyrus and the effec to exogenous bFG F injected in lateral ventricle afte r global cerebral ischemia -reperfu sion in rats.Results The level of neurogenesis in the dent ate gyrus of rats reached peak at 14d a fter global ischemia reperfusion.Not only the level of neurogenesis in the dentate gyrus in crease,but also the peak time of neur ogenesis was in advance after exogen ous bFGF injected in lateral ventric le.Conclusion Cerebral ischemia could induce the l evel of neurogenesis up-regulation.Exogenous bFGF could enhance the neurogenesis.
