摘要
目的 探讨维生素 C,E对家兔 60 Coγ线照射外周血的保护作用以及剂量效应关系 .方法 选择中国本兔作为研究对象 ,按质量性别均衡的原则随机分为 11组 ,每组 5只 ,自由摄食饮水 .实验第 1日开始各 Vit.C组进行 Vit.C针剂 iv,Vit.E组进行 Vit.E油剂 ig,阴性对照组与阳性对照组 ig相同量的脱除 Vit.C,Vit.E的花生油 .Vit.C的注射剂量分别为 10 ,2 0 ,40和 80 mg·kg- 1 ,Vit.E的灌胃量分别为 5 ,10 ,2 0 ,40和 80 mg· kg- 1 ,连续 5日 .喂养第 3日阳性对照组及各 Vit.C,Vit.E组的家兔进行 60 Coγ线照射 ,照射剂量为 4.5Gy.照后继续给药至第 5日 ,处理前 12 h停喂饲料 ,不停水 ,乙醚麻醉家兔 ,立即剖腹取腹腔静脉血 ,肝素抗凝后送检 ,分别测定外周血细胞数量及相关参数的变化 .结果 家兔经 4.5 Gy的 60 Co照射后 ,辐射各组实验动物的外周血白细胞(WBC)与对照组的白细胞数相比明显减低 (P<0 .0 1) ,维生素 C,E未表现出保护 WBC作用 ;外周血红细胞 (RBC)与对照组相比 ,除 Vit.C10 mg· kg- 1组和 Vit.E2 0 mg· kg- 1组具有一定的保护作用外 (均为 P<0 .0 5 ) ,其他各组均呈现下降趋势 ;血红蛋白 (HGB)与对照组相比 ,除 Vit.C10 m g·kg- 1组具有一定的保护作用外 (P<0 .0 5 ) ,其他各组也呈现下降趋势 ;
AIM To study the effects of vitamin C (Vit.C) and vitamin E(Vit.E) on peripheral blood composition and the dose effect relationship through the model of rabbit injured by radiation. METHODS Chinese rabbits were chosen as the experimental animals and were randomly divided into 11 groups, with 5 in each group. They were negative control group, positive group, trial groups of Vit.C, Vit.E groups containing four different dosage of Vit.C and five different dosage of Vit.E. Peanut oil was selected as diluent which was extracted by anhydrous alcohol for three times in same volume proportion of them, and then heated in 270 ℃ for two hours to delete residues of Vit.E and alcohol. The four Vit.C groups were received Vit.C iv with the dosages of 10, 20, 40 and 80 mg·kg -1 everyday respectively. The five Vit.E groups were received Vit.E ig with the dosage of 5, 10, 20, 40 and 80 mg·kg -1 everyday respectively. On the third day all groups except negative control group were radiated by 60 Co γ ray with the dose of 4.5 Gry. Then they were fed as before. On the sixth day all rabbits were killed and blood was taken for the analysis of peripheral blood composition changes by means of Auto machine. RESULTS After the radiation, the WBC numbers of peripheral blood in radiation groups were lower than those in the negative control group ( P <0.01). The RBC numbers of peripheral blood in 10 mg·kg -1 Vit.C supplemental group and 20 mg·kg -1 Vit.E supplemental group were higher than that in the positive control group ( P <0.05). The content of HGB in 10 mg·kg -1 Vit.C supplemental group was much higher than in both contrast groups of negative control group ( P <0.05) and positive control group ( P <0.05). The PLT numbers in Vit.E supplemental groups declined with the increase of Vit.E supplentation. The 10 mg·kg -1 Vit.C supplemental group showed some protective effects on the mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), mean platelet volume (MPV). CONCLUSION Lower dose of Vit.C supplementation had some protective effects on the peripheral blood composition of rabbits injuried by the radiation.
