柴胡及五灵丸对慢性肝损伤小鼠的影响

Effects of Chaihu and Wulingwan on chronic liver lesion in mice

目的 观察柴胡与五灵丸对慢性肝损伤的作用 .方法 采用四氯化碳 (CCl4)、D-氨基半乳糖 (D- Gla N)和脂多糖(L PS)与卡介苗 (BCG)多次注射造成 3种慢性肝损伤模型 ,给予同剂量的柴胡及五灵丸 ,测定血清转氨酶 (AL T、AST)、胆碱酯酶 (CHE) ,匀浆 AL T与 CHE、谷胱苷肽 (GSH)、丙二醛 (MDA)、碱性磷酸酶 (AL P)、一氧化氮 (NO)、白介素 2 (IL -2 )、白介素 6 (IL - 6 ) ,HE染色观察肝组织形态学 .结果 柴胡与五灵丸能有效地降低 CCl4所致慢性肝损伤小鼠血清与匀浆 AL T,升高血清 CHE,降低 GSH与 MDA,但无降瘀胆作用 ,五灵丸降瘀胆显著 (P<0 .0 1) ,HE显示受损的肝细胞有较好的恢复作用 .对 D- Gla N升高的血清 AL T有显著抑制作用 ,升高 CHE,降低 MDA,但不降 GSH,HE显示五灵丸修复损伤的肝组织不如柴胡 .两药对免疫性肝损伤升高的 AL T,AST有显著降低作用 (P<0 .0 1) ,使 IL - 2升高 ,柴胡抑制 IL -6与 NO,五灵丸显著升高 IL - 6与 NO.结论 两药对三种慢性肝损伤模型有不同程度的作用 ,对 CCl4所致慢性肝损伤五灵丸优于柴胡 ,对 D- Gla

AIM To observe the effect of Chaihu and Wulingwan on chronic liver injuries in mice. METHODS Chronichepatic injuries were established by injections of tetrachloromethane(CCl 4), D Galactosamine (D GlaN), lipopoly saccharide (LPS) +bacilli calmette guein (BCG) into mice respectively and them to which the same dosage of therapeutic medicines were administrated . Then levels of alanine aminotransferase (ALT) and cholinesterase in serum or homogenate were determined, level of Glutathione alkalinephosphatrase (ALP), molondiadehyde (MDA), interleukin2 (IL 2), interleukin6 (IL 6) were measured as well, the histopathologic changes were examined by HE staining. RESULTS Chaihu and Wulingwan significantly decreased ALT and elevated CHE for serum, and reduced GSH and MDA for homogenate in chronic liver injuries, Chaihu did not decrease the Glutathione in liver and Wulingwan cleared up them, meanwhile histopathologic results indicated effective recovery in chronic liver damage induced by D Glan. In contrast, Chaihu markedly inhibited the elevation of ALT in serum, while litered CHE and reduced MDA but did not decrease GSH in mice . Histopathological results displayed that effect of wulingwan was weaker than Chaihu in improving liver injuries. In addition, they both lowered ALT, AST markedly and elevated IL 2, but Chaihu inhibited IL 6 and NO and Wulingwan litered markedly IL 6 and NO. CONCLUSION Both have better cure effect on three kind chronic liver injuries by CCl 4, D GlaN immunirat. However, the effect of Wulingwan is better than that of Chaihu in mice chronic liver injuries by CCl4 and that of Wulingwan is weaker than that of Chaihu in lesion by D Glan.
