摘要
目的 探讨生长抑素类似物奥曲肽对胃癌细胞株MKN4 5生长的体外调节作用。方法 MTT法和细胞增殖核抗原 (PCNA)免疫细胞化学染色检测细胞增殖情况 ,RIA法测定表皮生长因子 (EGF)、转化生长因子 -α(TGF -α)的含量。结果 奥曲肽对胃癌细胞生长有抑制作用 ,以 1× 10 -6mol/L浓度最明显 ,并能抑制胃癌细胞合成EGF、TGF -α。结论 奥曲肽对胃癌生长具有负性调控作用 。
Objective To study the regulatory effects of somatostatin analogue(octreotide)on the growth of gastric cancer cell line MKN45 in vitro. Methods Cell proliferation was evaluated by means of MTT assay and PCNA immunocellularchemical staining. The contents of EGF and TGF-α were determined by radioimmunoassay (RIA). Results The growth of human gastric cancer cell line MKN45 was inhibited by octreotide, especially at 1×10 -6 mol/L, and the contents of EGF and TGF-α were also significantly inhibited (P<0.05). Conclusion Octreotide inhibits the growth of gastric cancer cells, which provides an experimental foundation for the use of ostreotide in the treatment of gastric cancer.
