摘要
目的 探讨阻断血小板源生长因子 (PDGF)B链表达对脑膜瘤细胞生长的影响。方法 在体外进行脑膜瘤细胞培养 ,观察转染PDGFB链反义寡核苷酸对脑膜瘤培养细胞生长的影响。结果 PDGFB链反义寡核苷酸能够阻断脑膜瘤培养细胞表达PDGFB链 ,转染后细胞增殖活性减弱 ,细胞生长受到抑制。
Objective To explore the feasibility of using platelet derioed growth factor(PDGF) B chain as a target for antisense gene therapy of human meningioma. Methods The human meningioma cell culture was conducted in vitro. An 18-mer antisense oligodeoxynucletide (AODN) complementary to a sequence starting at the ATG initiation codon of human c sis was synthesized and column purified. The cultured human meningioma cells were exposed to AODN. The effect of AODN on the growth of cultured meningioma cell was evaluated by MTT assay. Results Treatment with PDGFB chain AODN down regulated PDGFB chain mRNA expression and inhibited the growth of meningioma cell in culture. Conclusions PDGFB chain may be selected as target for gene therapy of meningiomas.
