甘露醇对大鼠局灶性脑缺血再灌注损伤时神经细胞凋亡的影响

Effect of mannitol on neuronal apoptosis in the focal cerebral ischemic reperfusive injury of rats

目的 研究甘露醇对大鼠局灶性脑缺血再灌注损伤时神经细胞凋亡的影响。方法 采用大鼠大脑中动脉阻断 (MCAO)模型 ,给予不同剂量甘露醇进行干预 ,用 TU NEL染色标记大鼠神经细胞凋亡情况 ,按 Swanson方法测量、计算梗死体积 ,并与未干预组及生理盐水对照组比较。结果 大、小剂量甘露醇均能减轻大鼠脑缺血再灌注后的神经细胞凋亡 ,并能阻止梗死体积的扩大。大、小剂量的效果之间无明显差异。结论 甘露醇的脑保护作用不完全与脱水降颅压有关 ,还可通过抑制神经细胞凋亡而发挥重要的脑保护作用。

Objective To study the effect of mannitol on neuronal apoptosis in focal cerebral ischemic reperfusive injury of rats.Methods Using TUNEL staining to label the condition of neuronal apoptosis in MCAO model rats, according to the way of Swanson,the infarction volume was measured and counted,and compared with the controls.Results Both big and small dose of mannitol could decrease the neuronal apoptosis in rats after cerebral ischemic reperfusion, and prevent the widening of the infarction volume.There was no significant difference between the effects of big and small dose of mannitol.Conclusion Mannitol injections can inhibit the neuronal apoptosis and play the role of brain's protection,the role doesn't link completely with its dehydration and reducing intracranial pressure.
