应用荧光原位杂交法和红细胞超氧化物歧化酶测定法诊断先天愚型的比较研究

摘要 目的 为探索快速诊断先天愚型症的方法 ,采用荧光原位杂交法与红细胞超氧化物歧化酶测定法进行研究。方法 红细胞超氧化物歧化酶用极谱氧传感器法测定 ,荧光原位杂交法按探针说明书进行。结果 红细胞SOD测定检测正常人 6 2例 ,正常新生儿 5 2例 ,先天愚型病人 32例 ,先天愚型新生儿 5例。SOD平均值分别为 :6 935± 487U/mlRBC、5 6 5 3± 46 3U/mlRBC、10 6 2 1± 837U/mlRBC、8375± 2 43U/mlRBC。荧光原位杂交法检测先天愚型病人 15例 ,正常人 15例 ,杂交斑点阳性率分别为 96 7%和 95 %。结论 红细胞超氧化物歧化酶测定法快速 ,可在 2h内完成 ;荧光原位杂交法相对费时、繁琐 ,但直观、安全 ,但两者的准确性是一致的。 Objective To explore the method of fluorescence in situ hybridization(FISH) and assay superoxide dismutase(SOD) of RBC to appraised Down Syndrome rapidly.Methods Testing SOD activity of RBC by the instrument of polarographic oxygen sensor;FISH by the direction of the reagent kit.Results The SOD average of 62 normal people was 6935±487U/mlRBC,52 normal newborns was 5653±463U/mlRBC,32 Down Syndrome was 10621±837U/mlRBC and 5 Down's newborns was 8375±234U/mlRBC.Assaied 15 Down's and 10 normal people by FISH,the percentage of hybridized spot was 96.7% and 95%.Conclusion Tested SOD of RBC was quickly,and finished in two hours.FISH would take more time and many steps,but it had clearly and objectly.Two method had a same veracity.
