显示人皮肤微血管构筑4种方法的实验研究 被引量:3

摘要 目的 :研究显示人皮肤微血管构筑的有效方法。方法 :采用酶组织化学的Gomori Takamatsu钙钴法和 (NBT/BCIP)硝基四唑氮蓝 / 5 溴 4 氯 3 吲哚磷酸盐 (NBT/BCIP)显色法 ,以及CD34和Ⅷ因子相关抗原的免疫组化法 ,显示人皮肤的微血管构筑。结果 :与传统方法比较 ,该 4种方法均能良好显示人皮肤的真皮内和真皮下的微血管构筑。免疫组化法的特异性好 ,但酶组织化学法通过厚切片技术 ,能更好显示微血管的网状结构。CD34的敏感性高于Ⅷ因子相关抗原。结论 :这 4种方法的初步确立 ,为皮肤和皮瓣的血运研究 ,甚至为瘢痕形成和创伤愈合等相关领域的研究 ,提供了可行的方法学依据。 Objective:To evaluate the effect of enzymic histochemical and immunohistochemical methods on showing microvascular architecture of human skin. Methods: Gomori Takamatsu method, NBT/BCIP method, CD34 and Ⅷ related antigen methods were used.Results:The four methods all were effective. Immunohistochemical methods were more specific than enzymic histochemical methods, but they were inferior to the latter, with the ability to demonstrate the network of microvessels. CD34 method was more sensible than Ⅷ related antigen method.Conclusion: Compared to traditional techniques, the four methods are all useful of the study on the microvascular architecture of human skin.
