上皮性卵巢癌组织β亚单位绒毛膜促性腺激素基因和蛋白表达

摘要 目的:探讨上皮性卵巢癌组织异位β-HCG基因和蛋白表达及其临床意义。方法:分别用逆转录聚合酶链反应(RT-PCR)、免疫组化(IHC)和放射免疫测定(RIA)方法检测上皮性卵巢癌细胞β-HCG基因和蛋白表达及癌组织培养上清和腹水中的β-HCG的浓度。结果:12例上皮性卵巢癌β-HCG基因表达全部阳性(100%)。7例免疫组化β-HCG蛋白阳性(58.3%)。培养上清和腹水中β-HCG平均浓度分别为0.586U/L和1.638U/L。结论:上皮性卵巢癌组织异位β-HCG的表达频率很高,β-HCG可以作为该肿瘤的标记物,RT-PCR方法的特异性,敏感性较高。 Objective:To investigate the ectopic expression ofβ-HCG gene and protein in ep ithelial ovarian cancer tissues and to evaluate its clinical significance.Methods:β-HCG gene transcription and translation were detected by reverse transcription polymerase chain reaction(RT-PCR)and immunohistochemical technique(IHC)in12cases of epithelial ovarian cancer tissues obtained from surgical operation.The concentration ofβ-HCG was measured in culture supernatants of all tumor tissues and in ascites from patients with ovarian cancer by radioimmunity assay(RIA).Results:β-HCG gene expression was positive in12/12ovarian cancer(100%).The protein ofβ-HCG was found in7/12ovarian cancer(58.3%).The mean concentration ofβ-HCG in culture supernatant and ascites of ovarian cancer pastients were0.586IU/L and1.638IU/L respectively.Conclusion:β-HCG is an useful tumor marker of ovarian cancer and a high frequent ectopic expression ofβ-HCG in epithelial ovarian cancer tissue can be detected by RT-PCR.
