摘要
目的 :研究As2 O3 对小鼠膀胱癌细胞系WYH92 9肿瘤细胞内游离钙离子浓度的影响。 方法 :用钙离子荧光探剂Fura 2 AM ,以荧光分光光度计检测As2 O3 对膀胱癌细胞内游离钙离子浓度的变化。 结果 :As2 O3 可以使肿瘤细胞 [Ca2 + ]i水平明显升高 ,由 88nmol/L逐渐升高至 2 30nmol/L ,存在明显的量效关系 (P <0 .0 5 ) ,而与细胞外钙离子浓度变化无关 (P >0 .0 5 )。结论 :As2 O3 影响小鼠膀胱癌细胞株WYH92 9肿瘤细胞内钙稳态 ,诱导肿瘤细胞内钙离子浓度持续升高 ;这可能是其抗肿瘤 。
Objective:To investigate the effect of arsenic trioxide(As 2O 3)on intracellular calcium concentration of mouse bladder transitional cell carcinoma cell line WYH 929 Methods:Tumor cells were treated with As 2O 3 at different mass concentration in vitro,probed with Fura 2-AM,the intracellular calcium concentrations were detected by fluometry.Results:The intracellular calcium concentration(i)was markedly increased by treatment with arsenic trioxide(P<0.05),and this increase was dose dependent,the i increase showed no relationship to the extracellular calcium concentration(P<0.05).Conclusion:As 2O 3 can markedly alter the intracellular calcium homeostasis of the mouse bladder carcinoma cell line WYH 929,which might be involved in its antitumor mechanism.
