摘要 目的 :建立毛细管区带电泳测定地西泮血药浓度的方法 ,并用该方法分析地西泮在大鼠体内的代谢情况。方法 :0 .0 5mol·L-1pH2 .5磷酸缓冲液为运行缓冲液 ,石英毛细管 5 0cm× 5 0 μmid ,紫外检测器 ,检测波长 2 14nm ,电动法进样 ,运行电压 15kV。结果 :地西泮在 10~ 10 0 0 μg·ml-1范围内线性关系良好 ,r=0 .9986 ,具有良好的精密度和准确度 ,测得地西泮在大鼠体内的消除半衰期约 2 .2 0h。结论 :高效毛细管区带电泳是一种进行药物分析的好方法 ,以其高效、快速、简便等特点 ,在药物分析中起着重要的作用。 OBJECTIVE The method of the measurement of the diazepam blood concentration by capillary zone electrophoresis was established and used to analyze the metablism of diazepam in rats.METHODS The running buffer was 0.05 mol·L -1 ,pH 2.5 phosphate buffer and the running voltage was 15 kV with electrokinetic injection, 214 nm UV detection wavelength.RESULTS The linearity of diazepam was 10~1000 μg·ml -1 , r= 0.9986 ,and the precision and accuracy was satisfied.The elimination half-life of diazepam in rats was 2.20 h.CONCLUSIONS Capillary zone electrophoresis is one of the best techniques in drug analysis field and gradually become very important for its′ high-efficiency?fast and good resolution simple.
