摘要
目的 探讨中国汉族人群SA基因微卫星的频率分布特征 ,明确SA基因座是否与中国汉族人原发性高血压 (EH)连锁。方法 选择上海地区汉族人 96例随机个体和 80对有EH家族史的EH同胞对 ,自外周静脉血白细胞中抽提DNA ,应用荧光标记PCR SSCP技术对SA基因微卫星的频率分布进行研究 ,以SA基因微卫星为遗传标记 ,通过状态一致性受累同胞对连锁分析确定SA基因座是否与中国汉族人EH连锁。结果 中国汉族人群SA基因微卫星的频率分布具有高度多态性 (多态信息含量PIC =0 .86 ,杂合度H =0 .88) ,但SA基因座与中国汉族人EH之间的连锁远未达到显著水平 (连锁分析值T =0 .972 ,P =0 .3847)。结论 SA基因座不与中国汉族人EH连锁 ,在SA基因座或其附近可能不存在中国汉族人EH易感基因。
Objective To investigate the frequency distributions of SA gene microsatellite (GT)n in Han Chinese, so as to ascertain the linkage of SA gene locus to essential hypertension in affected Han Chinese. Methods Among Han Chinese in Shanghai,96 individuals selected randomly and 80 essentially hypertensive sibpairs with such family history were collected. Fluorence marked PCR SSCP technique was applied to detect the frequency distributions of SA gene microsatellite (GT)n in Han Chinese. The linkage of SA gene locus to essential hypertension in Han Chinese was analyzed by Green's identity by status for affected siblings. Results the frequency distribution of SA gene microsatellite (GT)n in Han Chinese was of high polymorphism(PIC=0.86,H=0.88),but SA gene locus was not linked to essential hypertension in Han Chinese( T=0.972, P =0.3847). Conclusion There might be no essential hypertension related gene susceptible to Han Chinese in or near SA gene locus.
