摘要
目的 :探讨利用人卵细胞膜抗原制备抗受精抗体的可能性。 方法 :半纯化人卵细胞膜 ,利用生物素标记监测纯化技术 ,经小鼠足部皮下注射免疫获得免疫血清后 ,采用间接免疫荧光标记及共聚焦显微镜检测其特异性 ,体外受精抑制试验观察其受精抑制能力。 结果 :获得的免疫血清对人卵细胞具有特异识别能力 ,其受精抑制作用与浓度呈正相关。 结论 :利用人卵细胞膜可获得特异的抗受精抗体。
Objective: To study the possibility for the recovery of oocyte plasma membrane antigen as to produce anti fertilization antibodies. Methods: First, recovery of oocyte plasma membrane antigen was done by frozen thawing lysis and ultracentrifugation; Then, immune serum was produced in the mouse; Finally, the specificity of immune serum was determined by indirect immunofluorescence, and its anti fertilization ability was verified by in vitro human gamete fusion test. Results: Immune serum against human oolemma could be raised in the mouse. This immune serum specifically recognized the human oocyte plasma membrane. The fusion of human gamete was inhibited in a concentration dependent manner by this immune serum. Conclusion: The method can be extended to monoclonal antibodies (mAb) production and may be a good strategy in the research of human immuno contraception.
