用基因表达谱芯片研究人正常胃和胃癌中与发育相关基因的价值 被引量:4

The study of growth associated genes in gastric cancer using cDNA microarray

摘要 用 8 46 4条人 c DNA制成表达谱芯片 ;利用胃和胃癌组织的 m RNA,通过逆转录方法将 Cy3和 Cy5两种荧光分别标记到两组 c DNA上 ;利用这种 c DNA探针与表达谱芯片进行杂交后扫描 ,通过计算机分析判定基因是否在上述组织中存在差异表达。结果示在 197条与生长发育相关的基因中 ,存在差异表达的共 10条 ,上调的8条 (0 .0 95 % ) ,下调的 2条 (0 .0 2 4% )。应用这种方法识别出的基因对胃癌的诊断和治疗具有重要的潜在价值。 To research the growth associated expressed genes in gastric cancer,DNA microarray were made by spotting PCR products of 8464 human cDNAs.The probes were prepared by labeling gastric cancer tissue mRNA and normal gastric mucosa mRNA with Cy5 dUTP and Cy3 dUTP separatedly through reverse transcription.The mixed probes were hybridized to the cDNA microarray, the fluorescent signals were scanned and showed the difference between 2 tissues.Among the 197 target genes,10 growth assocoated differentially expression genes exist,8 genes were up(0.095%) and 2 genes down(0.024%).These approaches are potential candidates for gastric cancer diagnosis and therapy.
