Determination of Tobramycin by HPLC With Precolumn Derivatization

方法 对妥布霉素HPLC测定方法进行了研究。目 的 选用2，4- 二硝基氟苯(DNFB)柱前衍生化，以Agilent Hypersil ODS为色谱柱，0.25%三羟甲基氨基甲烷溶液-乙腈-0.5mol·L-1硫酸溶液(40∶59∶1)为流动相，流速为1.2ml·min -1，检测波长365nm。结果 在10～100μg范围内峰面积与浓度呈良好的线性关系，r＝0.9999。平均回收率为100.9%，RSD为1.5%。结论 本方法简便准确，重 现性好，可用于妥布霉素的质量控制。

OBJCTIVE To determine of tobramycin by MPLC. METHOD The Pre-column derivetization using 2,4- dinitrofluorobenzene(DNFB) Was Carried out. The separation was perfomed on a Agilent Hypersil ODS column. The mobile phase was 0.25% tris(hydroxymethyl)aminomethane-acetonitrile 0.5mol·L-1 sulfuric acid (40∶59∶1). The flow rate was 1.2ml/min and detected at 365nm. RESUITS The linear responsed in the ranged of 10-100μg(r＝0.9999).The average recovery was100.9%(RSD＝1.5%). Conclusion:The method wis simple, accurate and highly reproductive, and can be used for the quality control of tobramycin.
