摘要
目的 研究灵芝多糖 (GLB7)对小鼠T细胞三磷酸肌醇 (IP3 )和二酰基甘油 (DAG)的作用 ,确定其对T细胞信号转导途径。方法 采用细胞培养、放射免疫分析、离子交换层析和薄层色谱法测定小鼠脾脏T细胞中IP3 和DAG的变化。结果 GLB7引起小鼠T细胞中IP3和DAG浓度升高。IP3 的达峰时间为 30s ,百日咳毒素 (PTX)对此现象有抑制作用 ;DAG的合成出现两个峰 ,第一个峰迅速短暂 ,峰值位于 30s,第二个峰是持续时间较长的迟发峰。GLB7对活化的T细胞中IP3 和DAG未产生明显影响。结论 IP3 /Ca2 +
OBJECTIVE To investigate the effects of GLB 7 on inositol trisphosphate(IP 3)and diacylglycerol(DAG) in murine T cell and to provide a scientific evidence for its immunomodulatory mechanism.METHOD Cell culture,radio-immunological asssy, anion-exchange columns and thin-layer chromatography were used.RESULT GLB 7 increased the production of IP 3 and DAG in mice.The peak of IP 3 was approached at 30 s.Pertussis toxin(PTX) inhibited GLB 7 from increasing the production of IP 3.In the production of DAG,two peak were found,the first peak was rapid and transitory, and the second was delayed. GLB 7 didnt influence the production of IP 3 and DAG in active T cells.CONCLUSION The both paths of signal transduction of IP 3/Ca 2+ and DAG/PKC were involved in the immunomodulatory of GLB 7 to T cells.
