摘要
目的 探讨大鼠胃黏膜癌变发生发展过程中bcl 2基因表达及其与端粒酶活性的关系。方法 采用MNNG、10 %NaCl及 4 0 %乙醇灌胃建立了大鼠胃黏膜癌变模型 ,分别采用原位杂交技术及免疫组织化学方法检测大鼠胃黏膜bcl 2mRNA、蛋白表达 ,采用端粒微孔板杂交法检测胃黏膜端粒酶活性。结果 本造模方法在大鼠胃黏膜诱发出浅表性胃炎 (chronicsuperficialgastritis,CSG)、萎缩性胃炎 (chronicatrophicgastritis,CAG)、肠化生 (intestinalmetaplasia,IM )、异型增生 (dysplasia ,Dys)、胃腺癌 (gastricadenocarcinoma ,GC)等病变。大鼠从正常胃黏膜发展到胃癌 ,bcl 2基因表达及端粒酶活性均逐渐增高 ,并且两者有高度的一致性 (P <0 .0 5 )。结论 大鼠胃黏膜癌变发生发展过程中 ,癌基因bcl
Purpose: To study the correlation of the expression of bcl-2 gene and telomerase activity. Methods: We detected expression of bcl-2 by using in situ hybridization and immunohistochemistry assay, and tested telomerase activity by using telomeric repeat amplification protocol (TRAP)-microtiter plate hybridization assay in rats of two groups: control, model in gastric cancer rats induced by N-methyl-N′-nitro-N-nitrosoguanidine (MN-NG), 10% sodium chloride and 40% ethanl. Results: Models of various phase of carcinomatous change of gastic mucosa in Wistar rats were estabalished, chronic superficial gastritis (CSG), chronic atrophic gastritis (CAG), intestinal metaplasia (IM), dysplasia (Dys), and gastric adenocarcinoma (GC). With the development of gastric cancer, the expression of bcl-2 gene and telomerase activity were increased by degrees, and, the former has an important correlation with the latter (P < 0.05). Conclusions: Telomerase activity has an important relation with expression of bcl-2 in the process of carcinomatous change of gastric mucosa in Wistar rats.
