再灌注致大鼠心肌生物膜损伤及益心康胶囊的保护作用 被引量:2

摘要 目的 :观察再灌注对心肌生物膜的损伤 ,评价中药复方—益心康胶囊 (H30 3)对生物膜保护作用。方法 :利用离体大鼠全心停灌 /再灌 (I/R)模型 ,预先灌注H30 3(0 .2 ,0 .1mg/mL ,灌注 10min) ,观察对MDA、磷脂酶A2 、磷脂、游离脂肪酸、膜脂流动性、ATPase及线粒体呼吸功能的影响。结果 :I/R可导致离体大鼠心脏生物膜产生严重损伤。H30 3可通过减轻大鼠心肌组织脂质过氧化程度、抑制PLA2 活性、升高线粒体磷脂含量、降低FFA水平、改善膜脂流动性、保护线粒体Ca2 + ATPase及肌纤维膜的Na+ ,K+ ATPase活性及改善线粒体呼吸功能等作用而发挥抗损作用。结论 :H30 Objective: The injury effect of reperfusion on myocardium bio membrane was observed and the protective effect of traditional Chinese medicine—Yixinkang capsule(H303) on bio membrane was evaluated. Methods: By using the model of global heart ischemia/reperfusion(I/R,20min/20min) in rats in vitro , perfusing H303(0.2,0.1mg/ml,10min) beforehand, we measured the changes of MDA, PLA 2, PL, FFA, LFU, Ca 2+ ATPase, Na +,K + ATPase and respiratory function of mitochondria. Results: The results showed that I/R induced significant injury of bio membrane of rat myocardium. The anti injury effects of H303 were supported by alleviating lipid peroxidation of rat myocardium, inhibiting the activity of PLA 2, increasing and drcreasing the content of PL and FFA respectively, improving membrane lipid fluidity, protecting the activities of Ca 2+ ATPase and Na +,K + ATPase, ameliorating the respiratory function of mitochondria. Conclusion: H303 possessed the protective effect on reperfusion injury of rat heart in vitro.
