摘要
目的 研究拟获得快速高效的β—NFG(β -神经生长因子 )纯化方法 ,并为其生物特性研究和临床应用奠定基础。方法 通过低温高速离子、离子交换层析、透析等过程 ,从人胎脑和SD大鼠脑中分离纯化 β -NGF ,用SDS -PAGE分析其分子量和纯度 ,并经PC12细胞突起生长试验鉴定其生物学活性。结果 从人胎脑和SD大鼠脑中分离纯化得到的 β -NGF提取物在SDS -PAGE图上均呈单一条带 ,它们都能很好地促进PC12细胞长出突起 ,且人胎脑β -NGF提取物促进PC12细胞长出突起的能力和人重组 β -NGF差别无显著性。 结论 实验获得了纯度高且生物活性良好的 β -NGF。在分离纯化的其他参数不变时 ,分离纯化过程的总时间和温度对 β -NGF的得率和生物学活性有明显影响。
Objective To study its biological characteristics and apply its clinically in future and establish a quick efficient method of β-NGF extraction.Methods β-NGF was extracted from human fetal brain and SD rat brains by low-temperature and high speed centrifugation,ion-exchange chromatography and dialysis.Its bioactivity was identified by neurite outgrowth test of PC12 cells,and its molecualr weight and purity was analyzed by SDS-PAGF.Results β-NGF extracted from human fetal brain and SD rat brains showed a single band on SDS-PAGE.It can promotes neurite outgrowth of PC12 cells very well without difference in β-NGF.Conclusion The extracted β-NGF has high biological activity and purity.In isolation and purification,the time and temperature have impostant affect on the biological activity and the obtainable rate of extration.
