5种造血生长因子对造血祖细胞集落形成的影响

摘要 目的 探讨IL - 3、IL - 6、GM -CSF、G2、PIXY32 1对骨髓造血祖细胞集落形成的影响。方法  5种造血生长因子分别采用 10 0、2 5 0、5 0 0、75 0、10 0 0u/ml浓度 ,进行骨髓造血祖细胞集落培养 ,从中找出峰值浓度 ,然后在峰值浓度下进行集落培养 ,计数不同造血生长因子条件下所形成的集落。结果 IL - 3、IL - 6、GM -CSF、G2、PIXY32 1对造血祖细胞集落形成的峰值浓度分别为 10 0、10 0、2 5 0、75 0、2 5 0u/ml。峰值浓度下 ,对CFU -E、BFU -Em、BFU -Eim、CFU -GM形成的作用强度不同 ,PIXY32 1条件下所形成的集落数最多。结论 这五种造血生长因子对骨髓造血祖细胞的集落形成均有促进作用 ,PIXY32 1效果显著。 Objective To study the effect of IL-3,IL-6,GM-CSF,G2,PIXY321 on formation of colonies of hematopoietic progenitor cells (HPCs). Methods At first, the culture of colonies of HPCs in human bone marrows was performed at the concentrations of 100,250,500,750,1000 u/ml of the five hematopoietic growth factors (HGFs). And then, at the different concentrations of the HGFs,HPCs were cultured and the colonies of CFU-E,BFU-Em,BFU-Eim,CFU-GM were counted.Results The peak concentrations of IL-3, IL-6,GM-CSF,G2,PIXY321 were 100,100,250,750,250 u/ml for peak formation.PIXY321 was more effective for colony formation of HPCs than other HGFs.Conclusion All the five HGFs were capable of increasing the number of HPCs and the effect of PIXY321 was more encouraging than that of other HGFs.
