摘要
本文参照Guth介绍的酸碱预孵育肌球蛋白ATP酶法并加以改进,使之避免了肌纤维易断裂、组织块内产生冰晶、缓冲液种类过多等现象。结果表明,改进后的方法稳定可靠,在切片上能清楚显示不同类型的肌纤维。
The myosin ATPase method used by Guth has been modified. The modification included soiling the small muscle block (7mm×3mm) with talcum powder and stand it in agar instead of immersing it directly in liquid nitrogen to avoid fracture of muscle fiber during sectioning and to protect the tissue from absorbing moisture in the air which produces ice crystals and in turn damages the tissue. Other managements,such as using glycine NaOH buffer which can maintain the required pH and certain buffer action,may simplify the procedure without changing several kinds of buffer. That intact muscle fiber types are seen in the sections has proved the modified method is reliable.
