人工胶原膜对大鼠皮肤创伤促愈作用的研究 被引量:4

摘要 目的 :观察人工胶原膜 (代号rackderm)对皮肤创伤创面的作用及其机制 ,为进一步将其作为人工皮肤支架提供试验依据。方法 :扫描电镜观察rackderm的表面结构 ,应用图像分析技术测量孔径大小 ;制备动物背部创伤模型 ,分别应用人工胶原膜和油纱布包扎创面 ,分阶段取材后 ,宏观观察伤口愈合情况 ,并应用病理学手段 ,结合图像分析观察rackderm的生物学作用。结果 :扫描电镜显示rackderm为多孔结构 ,孔径介于 2 0~ 4 0 μm ,允许成纤维细胞和毛细血管的长入 ;宏观观察rackderm组伤口愈合时间明显早于油纱布组 ,组织学观察显示该组肉芽组织含量明显多于油纱布组 ,免疫组织化学染色显示促进伤口愈合的生长因子在该组表达明显增强。结论 :rackderm初步满足作为人工皮肤支架的物理条件 ,作为人工敷料 ,对创伤后造成的皮肤缺损 ,具有促进伤口愈合的作用。 Objective:To observe the effect of collagen membrane on healing of skin wounds and its mechanism in order to provide the experimental basis for its use as the trestle of artificial skin.Methods:Scanning electron microscope (SEM) was used to observe the surface structure of the membrane and the image analysis system was used to measure the diameter of the aperture of the membrane.Animal models of skin wound were set up.Half of the wounds were bound up with rackderm and the others were with vaseline gauze, then the wounds were observed grossly and under microscope.Results:The diameter of the apertures was between 20 to 40 microns and the fibroblasts and capillaries were able to grow into the rackderm.The grafting experimental results showed that after the rats were injured, the wounds bounded up with the collagen membrane healed earlier than that with the vaseline gauze, the granulation tissue in the rackderm dressed wound was also more abundant than that in the other group and so was the expression of growth factors. Conclusions: Collagen dressing can promote the healing of skin wound,and may be a satisfactory trestle for the tissue engineering of skin.
