摘要 利用阴离子交换色谱和疏水相互作用色谱从烟草疫霉菌培养液中分离出一种新的31ku激发子蛋白 ,选择了阴离子交换色谱流动相的最佳 pH值6.0 ,建立了疏水相互作用色谱硫酸铵Tris缓冲液 -水洗脱模式 ,简化了纯化步骤和减少了活性损失的危险。 A new 31 ku elicitor from a culture medium of P.parasitica Var. nicotiana was separated and purified by a chromatographic procedure including anion exchange chromatography and hydrophobic interaction chromatography. The chromatographic conditions were studied. The optimum pH value was found to be 6.0 for anion exchange chromatography. An elution mode of Tris buffer of ammonia sulfate-water was established for hydrophobic interaction chromatography. The procedure simplified working process and decreased the activity loss of the elicitor.
