摘要
目的 建立血清总胆汁酸 (TBA)测定的酶循环法。方法 基于用脱氢酶 辅酶体系循环底物的原理 ,建立了本法。结果 本法灵敏度 5 0 μmol/L ,A40 5nm ,1cm>0 .15 ;线性达 180 μmol/L ;批内CV <1.87% ,日间CV <2 .94 % ,总CV <3 .64% ;与普通酶显色法比较 :Y =0 .92 1X - 1.4 2 6,r=0 .9982 ,n =64。胆红素 <85 0 μmol/L、血红蛋白 <5 g/L、抗坏血酸 <2 .84mmol/L、乳酸 <2 4mmol/L、乳酸脱氢酶 <10 0 0 0U/L时 ,对结果无显著干扰。结论 本法灵敏度高 ,线性、精密度好 ,几乎无干扰 ,是一种理想的TBA测定方法。
Objective A method of the enzymatic cycling assay for total bile acids in serum was estabilished.Methods This method was established,based on the principle of the substrate cycling,dehydrogenase coenzyme system.Results The sensitivity was(△ A 405nm,1cm )>0 15 for 50 μmol/L TBA;The linearity was up to 180 μmol/L,with run CV <1 87%,between run CV <2 94%,and total CV <3 64%.The new assay( Y )was compared to the commercial enzymatic method( X ): Y =0 921 X -1 426, r =0 9982, n =64.No interference were showed when addition of bilirubin<850 μmol/L,Hb<5 g/L,ascorbic acid<2 84 mmol/L,lactic acid<24 mmol/L,LDH<10 000 U/L to a sample which TBA concentration was 112 7 μmol/L,respectively.Conclusions The sensitivity of this method was high.The linearity and precisions were good.No interference happened in determination of TBA in serum.
