摘要
通过正交实验确定了水解琼胶的最佳方案为盐酸浓度 0 .0 5 mol· L-1、水解 90 min、底物浓度 4 % ,此条件下二糖得率为 4 0 %。水解产物经乙醇沉淀、超滤除去大分子后 ,经凝胶柱层析可得到琼胶低聚糖各寡糖组份。用化学发光分析法比较了琼胶、琼脂糖水解液在抗坏血酸 - Cu2 + -酵母悬浮液 -鲁米诺 - H2 O2 体系中清除羟基自由基 (· OH)的能力 ,两者清除· OH的 IC50 分别为 0 .0 5 g· L-1和 0 .313g· L-1。
The optimum condition for hydrolysis of agar with hydrochloric acid was determined by the method of orthogonal experiments.The percentage of agarobiose yield reached 40% when the hydrolysis was conducted with 4% substrate at 0.05 mol·L -1 hydrochloric acid for 90 minutes.The hydrolytic product was fractionated by ethanol precipitation and ultrafiltration,and further separation was achieved by gel filtration.The ability of scavenging hydroxyl radical by agar and agarose hydrolytic products was also determined,and their IC 50 are 0.05 g·L -1 and 0.313 g·L -1 respectively.
