摘要
目的 :探讨糖尿病肾病系膜细胞功能改变及意义。方法 :采用STZ复制糖尿病模型 ,取肾进行系膜细胞培养 ,3 H -TdR法测定细胞增殖 ,用fluro - 3探针经共聚焦显微镜检测 [Ca2 + ]i 变化 ,ELISA法测定纤维连接蛋白 (FN)分泌量。结果 :糖尿病大鼠系膜细胞3 H -TdR掺入率和FN分泌量较正常大鼠增加 ,基础 [Ca2 + ]i 两组无差异 ,但糖尿病系膜细胞 [Ca2 + ]i 对血管紧张素II反应明显减弱。结论 :糖尿病系膜细胞增殖和分泌FN增加参与了糖尿病肾病细胞外基质的积聚 ,其 [Ca2 +
Objective: To explore the function modulation of mesangial cells in diabetic nephropathy. Methods: Diabetic mesangial cells (DMC) were cultured from streptozotocin induced diabetic rats. Cellular proliferation and fibronectin (FN) production were analyzed by 3 H-thymidine incorporation and ELISA respectively. Calcium levels ([Ca 2+ ] i) were measured using fluro-3 as flurophore by laser confocal microscopy. Results: 3 H-TdR incorporation and FN production from DMC were increased in contrast to the normal mesangial cells (NMC). Basal [Ca 2+ ] i in DMC was not different from that in NMC, but a blunted response to stimulation of the angiotensin II was observed. Conclusion: The results indicate that the phenotypic change of the mesangial cells occures in diabetes and these modulatiosn play a role in diabetic nephropathy.
