摘要
采用 He-Ne激光和化学诱变剂 Li Cl对少根根霉孢子和原生质体进行复合诱变 ,从一株产 GL A的出发菌株少根根霉 R8#中选育出高产菌株 RC3 78.摇瓶培养含油脂量 47.8% ,GLA含量 1 2 .6% ,比出发菌株油脂含量提高了 1 3 0 .8% ,GLA含量提高了 2 76.7% .从菌落形态、同工酶酶谱、红外光谱吸收特征和油脂产量及组成比较分析表明 :该菌株较出发菌株具有显著的变异 .菌株 RC3 78经传代培养 ,产脂量维持在 41 .1 %~ 48.7% ,GL A含量维持在 1 0 .8~ 1 2 .6% .
A mutant strain RC378 accumulating a large amount of lipid was successfully obtained when the original strain R8# was treated by He-Ne laser and LiCl as compound mutagen.The content of lipid in RC378 cultivated in shaking flask was up to 47.8%,which was 130.8% higher than that in the original strain.The content of GLA in Lipid from RC378 was 12.6%,which was 276.6% higher than that from R8#. R8# were analyzed and identified by morphological character,the isozyme patterns,infrared analyzed and identified by morpholohical character,the isozyme patterns,infrared absorption and lipid producing ability.The results proved the genetic materials of R8# had already changed after the mutation.After 10 times of transplantation,the content of lipid maintained at 41.1%~48.7%,GLA at 10.8%~12.6%.This showed that the genetic characteristics of mutant RC378 was stable.
