摘要
摘 要:目的 研究mdx鼠体内骨骼肌细胞凋亡情况,探讨凋亡对 Duchenne型肌营养不良症(DMD)的发生发展所起的作用。方法 采用流式细胞仪碘化丙啶(PI)排斥法测定不同渗透压下成肌细胞的凋亡数目,用TUNEL法测定不同鼠龄的mdx鼠骨骼肌细胞的凋亡情况。结果mdx鼠的成肌细胞在外界渗透压变化的情况下,发生凋亡的细胞数较C57鼠明显增加,且随mdx鼠鼠龄的增长而增加。结论 抗肌萎缩蛋白(dys)的缺失引起的凋亡在DMD疾病的启动和病程进展中可能起重要作用。
Abstract: Objective To investigate the apoptosis of skeletal muscles in mdx mice,and study the effect of apoptosis on the pathogenesis in Duchenne muscular dystrophy (DMD). Methods Apoptosis of myoblasts of C57 mice and mdx mice under different osmotic pressures was detected by flow cytometry (FCM) .and apoptosis of mdx mice skeletal muscles in different age was examined by TdT-mediated dUTP nick end labeling (TUNED. Results The number of apoptic mdx mice myoblasts is more than C57 mice when the external osmotic pressures are changed,and with the age increasing,apoptosis of skeletal muscles in mdx mice increases obviously. Conclusion Apoptosis may take a key role in pathogenesis and progression of DMD.
