健骨二仙丸含药血清对人成骨细胞增殖及细胞周期的影响

Effect of Jiangu Erxian Pill on Proliferation and Cell Cycle of Human Osteoblast

目的 :在临床研究和动物实验的基础上 ,进一步探讨健骨二仙丸对成骨细胞增殖和细胞周期的影响。方法 :分离、培养人的原代成骨细胞 ,采用3 H 胸腺嘧啶掺入法和四氮唑蓝法检测细胞增殖 ,流式细胞仪检测细胞周期。结果 :健骨二仙丸中、高剂量组细胞增殖率、S期细胞比率和细胞增殖指数等均显著高于空白对照组 (P <0 0 1,P <0 0 5 ) ,与雌激素组比较差异无显著性。细胞凋亡率和G0 G1期细胞比率显著低于空白对照组 (P <0 0 1,P <0 0 5 )。结论 :健骨二仙丸能有效促进成骨细胞的增殖、分化 。

To further explore the effect of Jiangu Erxia n Pill (JGEXP) on proliferation and cell cycle of human osteoblast on the ba sis of previous clinical and experimental studies. Methods: Human primary osteoblast were isolated and cult ured. The cell proliferation was tested by 3?H thymine incorporation and met hyl thiazolyl tetrazolium (MMT) method and the cell cycle was determined by flow cy tometry technique. Results: In the medium and high dosage JGEXP groups, the cel l proliferation rate and index, and percentage of diploid synthesis phase (S phase ) cells were significantly higher than those in the blank control group ( P<0 01 or P<0 05), and similar to those in the estro gen group; and the cell apoptosis rate and percentage of G 0 G 1 stage cells were lower than those in the blank control group (P<0 01 and P<0 05). Conclusion: JGEXP could effectively promote the cell prolifera tion and differentiation, and prevent the cell apoptosis of osteoblast in vitro.
