摘要
目的 观察骨质疏松大鼠不同时期骨密度 (BMD)、血清胰岛素样生长因子 - 1 (IGF- 1 )的浓度以及转化生长因子 β1 (TGF- β1 )的骨表达 ,进一步探讨绝经后骨质疏松与细胞因子的相关性。方法 用摘除双侧卵巢的方法制备实验大鼠骨质疏松模型 ,实验组行摘除双侧卵巢术 ,对照组行假手术 ,仅切除卵巢周围的脂肪组织 ,应用 HOLOGIC第 4代双能 X线 450 0 w骨密度仪测量去卵巢不同时期 (术后 8w、1 2 w、1 6w)大鼠全身骨密度和腰椎感兴趣区域 (L1 ～ L4 )骨密度 ,并采用 EL ISA法和免疫组化的方法 ,检测去卵巢大鼠不同时期血清 IGF- 1水平和骨组织中 TGF-β1 表达。结果 从术后第 8周到第 1 6周去卵巢组腰椎骨密度均明显低于对照组 (P<0 .0 1 ) ,全身骨密度从术后第 1 6周开始降低。从术后第 8周到第 1 6周去卵巢组血清 IGF- 1浓度值均低于对照组 (P <0 .0 5) ,去卵巢组术后 1 2 w骨组织中成骨细胞、软骨细胞、骨基质 TGF-β1 表达降低。结论 绝经后骨质疏松的发病与 IGF- 1、TGF-β1
Objective To observe the bone mineral density(BMD), insulin like growth factor 1(IGF 1) in serum and transforming growth factor beta 1(TGF β1) in bone during different period of osteoporosis rat, and explore the relationship between postmenopausal osteoporosis and the growth factor Methods 60 female SD rats(12 month old) were randomized into two groups of 30 each, the experiment group and the control group The experimental model of osteoporosis was established by removing twin ovary operation Bone mineral densities of global and the lumber spine were measured with HoLoGEC dual energy X ray bone densitometer in rat Serum IGF 1 levels and bone TGF β1 expression during different period of osteoponosis rat were investigated by means of ELISA and immunohistochemichy respectively Results At 16th week after operation, the BMDs of global in the experiment group were lower than those in the control group(P<0 01) The BMD of the lumber spine in the experiment group was lower than that in the control group(P<0 01) at different periods The serum IGF 1 concentration was lower in the experiment group(P<0 05) compared to the control group during 8 to 16 weeks after operation The expressions of TGF β in chondrocytes and osteoblasts were different in the experimental group compared to the control group Conclusions TGF β1 and IGF 1 may be important in the pathogenesis of the postmenopausal osteoporosis
