摘要
本实验选用雌性Wistar大鼠,妊娠第一天腹腔注射四氧嘧啶(15mg/100g体重)。分别于妊娠第17、18、19、20天剖腹取胎胰,一部分用改良Bouin液固定,石腊包埋切片,HE染色及免疫组织化学PAP法显示β细胞,并测定β细胞的数面密度和体密度;另一部分常规电镜制片及观察。结果显示,实验组β细胞的数面密度均显著高于对照组,第17、18天实验组胎鼠β细胞分泌尿粒校对照组的增大,数量增多。而17天胎鼠胰岛为非常不规则的内分泌细胞团,对胰岛素抗血清呈阳性反应的细胞少,阳性反应弱。随着胎鼠的生长发育胰岛细胞逐渐聚集成条索状或团块状,阳性细胞增多,阳性反应渐增强。上述结果提示:妊娠高血糖可以促进胰岛β细胞数量增多,分泌颗粒成熟。
Female Wistar rats received intraperitoneal lnjection of a1loxan(15mg/100g weight)after pregnancy. The animals were killed on the 17, 18, 19, 20 day of pregnancy and the pancreas of fetuses were observed by light and electronic microscope. The islet 5 cells were showed by the immunohistochemistry (PAP method) and NA and Vv of 9 cell were measured. There were a definite rise for NA of 5 cells in fetuses of diabetic mother rats and their secretory granules were more and bigger than control group. In the meantime,the endocrine cells of pancreas gathered to form glomerulosa-like or fasciculata-like structure from 17 to 20 day of pregnan-cy. It is concluded that the maternal diabetes could accelerate the incfease of β cell number and the maturation of it's secretory granules.
