人参总皂甙诱导L—细胞产生造血调节因子的实验研究

摘要 为探讨中药人参的重要有效成分人参总皂甙(TotalSaponinsofPanaxGinseng,TSPG)调节造血的可能机制,在体外祖细胞培养中以TSPG诱导(LCCMTSPG)或未经诱导(LSCM)的L—细胞条件培养液替代祖细胞生长因子,观察集落产率的变化,并与TSPG和LcCM共同作用(LcLM+TSPG)后的集落产率作比较。结果如下:LcCMTSPG作用后的CFU-GM(位—单系祖细胞)、BFU-E(早期红系祖细胞)、CFU-E(晚期红系祖细胞)的集落产率分别为24.8±9.01、61.73±11.26、145.93±17.22,与LcCM组(51.85±10.08、33.95±825、108.66±13.56)相比分别为下降(P<0.05)、提高(P<0.01)、提高(P<0.01)。提示TSPG可诱导L—细胞产生选择性作用于粒—单系的造血抑制因子和作用于红系的生长因子。LCCM+TSPG作用后CFU-GM、BFU-E、CFU-E集落产率分别为74.25±10.67、42.5±9.32、98±10.81,与LcCM组相比分别为提高(P<0.05)、提高(P<0.05)、无变化,提示TSPG可能与LcCM中促进CFU-E、BFU—E生长的因子协同作用。该研究表明TSPG可经影响基质细胞产生造血调节因子或与造血调节因子共同调节血细胞发生。 For exploration the effect of thc main component of Chinese mediclnc Panaxginseng-to-tal saponins of Panaxginseng (TSPG) on regulation of hematopoiesis, wc uscd L-cell condi-tioned mcdium (LcCM ) induced by TSPG (LcCM.TSPG) to replacc exogencous hcmatopoiesisgrowth factor (HGF) in vitro hematopoietic progenitor culture. The result showcd: Thecolony number of CFU-GM in LcCMTSPG group decreased as compared with LcCM group,but that of BFU-E and CFU-E increased. The colony number of CFU-GM and BFU-Ein LcCM+TSPG group incrcased, as compared with that of LcCM group, but that of CFU-E was no significant different. The result suggests that TSPG may induce L-cell to se-cretc sclectioely CFU-GM inbibitor and,BFU-E and CFU-E growth factor; TSPG maycooperate also with the growth factors in LcCM of CFU-GM and BFU-E. It means thatTSPG may induce L -cell to produce hematopoiesis regulatory factor (HRF) or cooperatewith LcCM HRF to regulate hematopoiesis.
